G protein-coupled receptor 81 (GPR81) also known as Hydroxycarboxylic acid receptor 1 (HCAR1) plays an important role in lactate signaling. It has a molecular mass of approximately 40 kDa. GPR81 is expressed mainly in adipose tissue as well as in muscle and liver cells. The receptor responds to changes in lactate levels in these tissues and influences metabolic processes accordingly.
Biological function summary

GPR81 mediates lactate sensing and regulates lipolysis. It does not form part of a larger complex but functions on its own transmitting signals inside the cell upon lactate binding. This action reduces the breakdown of lipids by inhibiting the enzyme adenylate cyclase which in turn decreases intracellular cAMP levels. By limiting lipolysis GPR81 plays a role in energy balance and metabolic homeostasis.

Pathways

GPR81 integrates into energy regulation and metabolic pathways. It is significantly involved in the lactate and lipid metabolism pathway connecting with adenylate cyclase as a downstream effector. The inhibition of adenylate cyclase reduces cAMP synthesis affecting the Protein Kinase A (PKA) signaling pathway. This pathway links GPR81 to other receptors that modulate energy homeostasis such as GPR109A.

Research connects GPR81 to obesity and type 2 diabetes. By modulating lipolysis in adipose tissue GPR81 impacts fat storage and insulin sensitivity. The receptor's activity could also indirectly influence pathways involving insulin receptor substrate (IRS) proteins key players in glucose regulation. Understanding and targeting GPR81 may provide therapeutic advantages for managing these metabolic conditions.
