GPCR RDC1 also known as CXCR-7 is a G protein-coupled receptor that binds to chemokines such as CXCL12 and CXCL11. The receptor has a predicted molecular mass of approximately 40 kDa. It expresses widely in various tissues with high levels found in the heart liver kidney and brain. This receptor plays a role in the trafficking and localization of different cells often working alongside or complementing other chemokine receptors.
Biological function summary

GPCR RDC1/CXCR-7 takes part in complex cell signaling related to cell growth survival and migration. It is not usually part of a classic GPCR signaling pathway which sets it apart from other receptors in its family. Instead it can modulate pathways by forming heterodimers with other receptors such as CXCR4 affecting both ligand availability and receptor sensitivity. CXCR-7 can also function as a decoy receptor binding ligands without initiating a typical signaling cascade.

Pathways

The participation of GPCR RDC1/CXCR-7 is noted in chemokine-mediated pathways. Of particular importance is the SDF-1/CXCL12 signaling pathway which it shares with CXCR4. This pathway plays an important role in the regulation of stem cell migration and homing. CXCR-7 modulates the bioavailability of CXCL12 and influences how CXCR4 triggers downstream effects affecting cellular response to the external environment.

GPCR RDC1/CXCR-7 holds significance in cancer and cardiovascular diseases. In cancer overexpression of CXCR-7 associates with tumor growth and metastasis often in conjunction with the action of CXCR4. In cardiovascular conditions altered expression and functional shifts in CXCR-7 can lead to abnormal vascularization contributing to disease progression. Its role in these diseases makes CXCR-7 a valuable target for therapeutic interventions.

Atypical chemokine receptor that controls chemokine levels and localization via high-affinity chemokine binding that is uncoupled from classic ligand-driven signal transduction cascades, resulting instead in chemokine sequestration, degradation, or transcytosis. Also known as interceptor (internalizing receptor) or chemokine-scavenging receptor or chemokine decoy receptor. Acts as a receptor for chemokines CXCL11 and CXCL12/SDF1 (PubMed : 16107333, PubMed : 19255243, PubMed : 19380869, PubMed : 20161793, PubMed : 22300987).
