GPR83 also known as GIR or G-protein coupled receptor 83 is a member of the class A rhodopsin-like family of G-protein coupled receptors (GPCRs). This receptor has a mass of approximately 47 kDa. GPR83 is expressed in several tissues with significant expression in the brain particularly in the hypothalamus and to a lesser extent in the cortex and brainstem. It is also found in immune system tissues like the thymus and lymph nodes suggesting roles in both neurological and immune functions.
Biological function summary

GPR83 engages in the regulation of neuroendocrine signaling. There is evidence that it affects appetite control and energy homeostasis implying a role in metabolic processes. GPR83 does not function as part of a larger receptor complex but interacts with other GPCRs or intracellular proteins to transmit extracellular signals into cells. Its activity can modulate synaptic transmission and impact behaviors linked to feeding and stress response.

Pathways

Protein interactions involving GPR83 connect mainly to neuropeptide Y (NPY) signaling and the broader GPCR signaling pathways. The receptor modulates signaling by potentially influencing second messenger systems such as cAMP. Proteins related to these pathways include NPY a neuropeptide that affects energy balance and various G-protein subunits that mediate downstream effects following GPR83 activation.
