ab208777 was shown to specifically react with Progranulin in wild-type HAP1 cells as signal was lost in GRN (Progranulin) knockout cells. Wild-type and GRN (Granulin) knockout samples were subjected to SDS-PAGE. ab208777 and ab8245 (Mouse anti GAPDH loading control) were incubated overnight at 4°C at 1/2000 dilution and 1/10000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

ab208777 Anti-Progranulin antibody [EPR15864] was shown to specifically react with Granulin in wild-type HEK293T cells. Loss of signal was observed when knockout cell line ab266738 (knockout cell lysate ab257235) was used. Wild-type and Progranulin knockout samples were subjected to SDS-PAGE. ab208777 and Anti-GAPDH antibody [6C5] - Loading Control (ab8245) were incubated at room temperature for 2.5 hours at 1 in 1000 dilution and 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

ab208777 was shown to react with GRN in wild-type HEK293T cells in Western blot with loss of signal observed in GRN knockout cell line ab266738. Wild-type HEK293T and GRN knockout cell lysates were subjected to SDS-PAGE. Membranes were blocked in 5% milk in TBST for 1 hr before incubation with ab208777 overnight at 4 °C at a 1/1000 dilution. Blots were incubated with secondary antibodies at 0.2ug/mL before imaging.

Granulin also known as progranulin or GRN is a protein with a molecular mass of approximately 68 kDa. This protein is encoded by the GRN gene and is involved in various cellular processes. Granulin is expressed widely within the body particularly in epithelial cells immune cells and neurons. Researchers have developed methods like GRN ELISA and progranulin ELISA to quantitatively measure the levels of granulin in biological samples aiding in further understanding of its role in health and disease.
Biological function summary

Granulin functions as a growth factor involved in tissue repair cell proliferation and inflammation. It does not function as part of a larger protein complex but acts individually in exerting its effects on cells.
