Granuphilin also known as synaptotagmin-like protein 4 (SYTL4) serves as an important player in vesicle trafficking and fusion with its role as a Rab effector protein. It weighs approximately 85 kDa. This protein mainly expresses in insulin-secreting beta cells of the pancreas and in adipocytes. Granuphilin directly binds to Rab3 and Rab27 influencing the movement and docking of secretory granules on the plasma membrane.
Biological function summary

Granuphilin impacts insulin secretion by managing the docking of insulin granules. It is an essential regulator within the secretory pathway. Granuphilin does not work alone but instead associates with a complex of proteins including Rab27A to facilitate precise targeting and fusion of vesicles with cell membranes. This interaction impacts the exocytosis process vital for hormone release.

Granuphilin integrates into the insulin secretion pathway and the exocytosis pathway.
