Supplementary information

The protease Granzyme M also known as EC 3.4.21.79 plays a vital role in immune function. It belongs to the serine protease family and has a molecular mass of approximately 27 kDa. Granzyme M is expressed in natural killer (NK) cells and cytotoxic T lymphocytes. These immune cells utilize Granzyme M to induce apoptosis in target cells acting as a defense mechanism against infections and tumors.
Biological function summary

The protease Granzyme M contributes to the induction of cell death. Notably it does not form part of a larger complex and acts independently. Its primary function is to cleave intracellular substrates which leads to the disintegration of target cells' structural proteins and the ultimate demise of infected or cancerous cells. While other granzymes share similar roles Granzyme M shows distinct substrate specificity which provides diversity in immune responses.

Pathways

Granzyme M functions significantly within the immune system's apoptotic pathways. It interacts indirectly with proteins such as caspases other granzymes including Granzymes A and B influencing these cell death pathways. While it does not directly cleave caspases Granzyme M helps dismantle vital cellular components facilitating further activity of caspase proteins in apoptosis.

Granzyme M exhibits connections to cancer and viral infections. Tumor cells and virally-infected cells become targets of Granzyme M's cytotoxic actions. Studies show overlapping functions among granzymes in targeting malignant cells with Granzyme M complementing other granzymes' efforts in eliminating harmful cells. Additionally its activity links to proteins like perforin which assists in delivering granzymes into target cells and heightens the immune system's response against pathological conditions.
