Adapter protein which modulates coupling of a number of cell surface receptor kinases with specific signaling pathways. Binds to, and suppress signals from, activated receptors tyrosine kinases, including the insulin (INSR) and insulin-like growth factor (IGF1R) receptors. The inhibitory effect can be achieved by 2 mechanisms: interference with the signaling pathway and increased receptor degradation. Delays and reduces AKT1 phosphorylation in response to insulin stimulation. Blocks association between INSR and IRS1 and IRS2 and prevents insulin-stimulated IRS1 and IRS2 tyrosine phosphorylation. Recruits NEDD4 to IGF1R, leading to IGF1R ubiquitination, increased internalization and degradation by both the proteasomal and lysosomal pathways. May play a role in mediating insulin-stimulated ubiquitination of INSR, leading to proteasomal degradation. Negatively regulates Wnt signaling by interacting with LRP6 intracellular portion and interfering with the binding of AXIN1 to LRP6. Positive regulator of the KDR/VEGFR-2 signaling pathway. May inhibit NEDD4-mediated degradation of KDR/VEGFR-2.

The Growth Factor Receptor-Bound Protein 10 often referred to as GRB10 is an adaptor protein with a mass of approximately 66 kDa. This protein interacts with various receptor tyrosine kinases and other intracellular signaling molecules. GRB10 is expressed in many tissues but it shows higher expression in the brain muscle and liver. It plays a significant role as a modulator in cellular processes that involve signal transduction.
GRB10 interacts with receptor tyrosine kinases such as the insulin receptor and engages in regulating the insulin signaling pathway. It acts as part of a complex that influences cell growth and metabolism. GRB10 binds Via its SH2 domain to activated receptors such as IGF-1R altering downstream signaling and having an impact on growth and metabolic processes.
GRB10 is integral to the PI3K-AKT signaling pathway and the mTOR pathway. It negatively regulates these pathways by binding to receptors and altering downstream effects. GRB10 interacts with proteins such as IRS1 and mTORC1 affecting how cells respond to insulin and growth factors. Through these interactions it contributes to cellular metabolism and growth regulation.
Studies show that GRB10 has connections to conditions like cancer and type 2 diabetes. Alterations in GRB10's function or expression can lead to abnormal cellular signaling contributing to oncogenesis due to disrupted growth signals. In type 2 diabetes GRB10 affects insulin signaling pathways which is related to insulin resistance. These disorders show interconnection with proteins like IRS1 where GRB10 modulates pathways influencing disease states.
