Supplementary information

GRF2 also known as GTP-binding protein GRF2 plays a mechanical role as a guanine nucleotide exchange factor (GEF) for the Rho family of small GTPases. It has a molecular mass of approximately 94 kDa. This protein assists in the exchange of GDP for GTP therefore activating Rho GTPases. GRF2 is highly expressed in brain tissue specifically in neurons indicating a role in neurological processes.
Biological function summary

GRF2 functions as a critical regulator of signal transduction pathways. It forms part of signaling complexes that transmit signals from receptors on the cell surface to intracellular effectors. Through its GEF activity GRF2 influences cytoskeletal reorganization which impacts cell morphology motility and growth. It interacts with other proteins such as ELMO1 to facilitate cellular responses to external stimuli.

Pathways

GRF2 integrates into the MAPK/ERK pathway amplifying signals that trigger cellular proliferation and survival. GRF2 also links to the Rho/ROCK pathway regulating actin dynamics and cellular architecture. Within these pathways GRF2 functions in concert with proteins such as Ras and RhoA ensuring effective transmission and modulation of signals that govern various cellular activities.

GRF2 has relevance to neurological and cancer-related conditions. In the context of neurological disorders alterations in GRF2 expression or function can contribute to the pathophysiology of conditions like schizophrenia. In cancer GRF2 may influence tumor progression linking to proteins such as Ras which is often mutated in various cancers. Understanding GRF2's role provides insight into its potential as a therapeutic target for these diseases.

Target data

Guanine nucleotide-releasing protein that binds to SH3 domain of CRK and GRB2/ASH. Transduces signals from CRK to activate RAS. Involved in cell branching and adhesion mediated by BCAR1-CRK-RAPGEF1 signaling and activation of RAP1 (PubMed : 12432078). Plays a role in the establishment of basal endothelial barrier function. Plays a role in nerve growth factor (NGF)-induced sustained activation of Rap1 and neurite outgrowth.
