Transcription factor playing important roles in primary neurulation and in the differentiation of stratified epithelia of both ectodermal and endodermal origin (By similarity). Binds directly to the consensus DNA sequence 5'-AACCGGTT-3' acting as an activator and repressor on distinct target genes (PubMed:21081122, PubMed:25347468). xhibits functional redundancy with GRHL2 in epidermal morphogenetic events and epidermal wound repair (By similarity). Exhibits functional redundancy with GRHL2 in epidermal morphogenetic events and epidermal wound repair but is essential to form the epidermal barrier with TGM3 as critical direct target gene among others. Despite being dispensable during normal epidermal homeostasis in the adulthood, is again required for barrier repair after immune-mediated epidermal damage, regulates distinct gene batteries in embryonic epidermal differentiation and adult epidermal barrier reformation after injury. Plays unique and cooperative roles with GRHL2 in establishing distinct zones of primary neurulation. Essential for spinal closure, functions cooperatively with GRHL2 in closure 2 (forebrain/midbrain boundary) and posterior neuropore closure (By similarity). Also required for proper development of the oral periderm (PubMed:24360809). No genetic interaction with GRHL3, no functional cooperativity due to diverse target gene selectivity (PubMed:21081122).

GRHL3 also known as Grainyhead-like 3 functions mainly as a transcription factor. This protein weighs approximately 68 kDa and manages gene expression by binding to specific DNA sequences. GRHL3 is essential for keratinocyte differentiation and maintains epidermal integrity. The gene encoding GRHL3 shows expression mainly in epithelial tissues such as skin lungs and bladder.
Grainyhead-like 3 coordinates the development and maintenance of epithelial tissues. It plays a significant role in wound healing by promoting cell migration and adhesion. GRHL3 forms part of a regulatory complex interacting with other transcription factors to control target genes involved in skin barrier formation. Besides its regulation extends to processes like epithelial-mesenchymal transition.
The involvement of GRHL3 is important in the epidermal growth factor receptor (EGFR) signaling pathway and the Wnt signaling pathway. These pathways allow GRHL3 to influence cell proliferation and differentiation. The protein interacts with others like EGFR and beta-catenin in these contexts which shows its importance in skin physiology and development.
GRHL3 is associated with skin-related conditions including psoriasis and non-melanoma skin cancer. Mutations or dysregulation in GRHL3 expression link to aberrant skin cell proliferation and differentiation. In these contexts GRHL3 connections to proteins such as p53 and NF-kB have been identified indicating potential pathways for therapeutic targeting and intervention strategies.
