G protein-coupled receptor kinase 6 (GRK6) a member of the GRK family plays an important role in phosphorylating activated G protein-coupled receptors (GPCRs). GRK6 regulates receptor desensitization through this mechanism. It is also known as GPRK6. GRK6 has an approximate mass of 66 kDa. This kinase is expressed in various tissues including the brain heart and lymphoid organs contributing to its wide-ranging impact in physiological processes.
Biological function summary

GRK6 modulates signaling pathways by controlling the desensitization and internalization of GPCRs. The GRK6 protein localizes to the cellular membrane and operates as part of a larger signaling complex. This regulation process affects cellular responses like migration and immune reactions. GRK6 influences the reactivation and recycling of GPCRs impacting the duration and intensity of receptor-mediated signals in cells.

Pathways

GRK6 integrates into the GPCR signaling and chemokine signaling pathways. It directly modulates the activity of GPCRs contributing to cellular responses tied to these pathways. Related proteins such as beta-arrestins and G proteins also play significant roles in these interactions. GRK6 can phosphorylate GPCRs facilitating beta-arrestin binding which affects receptor trafficking and signaling.

Target data

Specifically phosphorylates the activated forms of G protein-coupled receptors. Such receptor phosphorylation initiates beta-arrestin-mediated receptor desensitization, internalization, and signaling events leading to their desensitization. Seems to be involved in the desensitization of D2-like dopamine receptors in striatum and chemokine receptor CXCR4 which is critical for CXCL12-induced cell chemotaxis (By similarity). Phosphorylates rhodopsin (RHO) (in vitro) and a non G-protein-coupled receptor : LRP6 during Wnt signaling (in vitro).
