Gamma-secretase activating protein (GSAP) is a critical regulator in the modulation of gamma-secretase activity influencing the production of amyloid beta-peptides. Also known by its alternative name GSAP the protein has a molecular mass of approximately 16 kDa. It is expressed in multiple tissues including the brain where its presence is significant due to its involvement in neurobiological processes.
Biological function summary

GSAP interacts mechanically with gamma-secretase and enhances its specificity for amyloid precursor protein (APP). This interaction plays a significant role in amyloidogenic processes and is part of a larger complex impacting the generation of amyloid beta-peptides. Through this interaction GSAP becomes an important factor in the regulation of pathways related to amyloid beta generation.

Pathways

GSAP actively participates in the amyloidogenic pathway which is pivotal in Alzheimer's disease pathology. In the context of this pathway GSAP closely interacts with gamma-secretase complex components including presenilins which significantly influence beta-amyloid levels. Likewise GSAP's interaction with APP reflects on its integral role in modulating the proteolytic cascade that generates beta-amyloid peptides.

GSAP's role is most notably linked to Alzheimer's disease. Its contribution to the production of beta-amyloid peptides positions it centrally in the pathogenesis of Alzheimer's wherein these peptides aggregate to form neurotoxic plaques. The interaction with presenilin proteins further ties GSAP to familial forms of Alzheimer's disease reinforcing its potential as a therapeutic target in mitigating such neurodegenerative disorders.

Target data

Regulator of gamma-secretase activity, which specifically activates the production of amyloid-beta protein (amyloid-beta protein 40 and amyloid-beta protein 42), without affecting the cleavage of other gamma-secretase targets such has Notch. The gamma-secretase complex is an endoprotease complex that catalyzes the intramembrane cleavage of integral membrane proteins such as Notch receptors and APP (amyloid-beta precursor protein). Specifically promotes the gamma-cleavage of APP CTF-alpha (also named APP-CTF) by the gamma-secretase complex to generate amyloid-beta, while it reduces the epsilon-cleavage of APP CTF-alpha, leading to a low production of AICD.
