The GSDMC protein also known as Gasdermin C possesses significant mechanical functions in cells. It has a mass around 54 kDa. The GSDMC protein is mainly expressed in epithelial tissues and can localize to the cytoplasm. Gasdermin C has a role in forming pores in the cell membrane which is a critical step during the pyroptotic cell death process. The protein's expression levels and mechanical properties influence its function in cells.
Biological function summary

GSDMC plays a role in the process of regulated cell death known as pyroptosis. This protein is part of the larger gasdermin family which affect the cellular membrane by creating pores. Through this pore formation GSDMC leads to the release of inflammatory cytokines. By modulating membrane integrity it contributes to immune responses and cell signaling which highlights its influence on cell viability and death.

Pathways

Studies show that GSDMC is involved in the inflammasome pathway and the TNF signaling pathway. It interacts with other proteins like caspase-1 which is important for activating pyroptosis and mediating inflammation. This participation in inflammasome activation positions GSDMC as an important regulator in immune response pathways and cellular homeostasis especially under inflammatory conditions.

Researchers have connected GSDMC with cancer progression and inflammatory diseases. The protein's aberrant expression is linked to colorectal cancer and Crohn's disease where it may alter cellular dynamics and impact disease pathology. It associates with proteins such as IL-1β in the context of inflammation which can influence the development and severity of these diseases making GSDMC a potential target for therapeutic intervention.

Gasdermin-C. This form constitutes the precursor of the pore-forming protein : upon cleavage, the released N-terminal moiety (Gasdermin-C, N-terminal) binds to membranes and forms pores, triggering pyroptosis.. Gasdermin-C, N-terminal. Pore-forming protein that causes membrane permeabilization and pyroptosis. Produced by the cleavage of gasdermin-C by caspase CASP8 in response to death signals. After cleavage, moves to the plasma membrane where it strongly binds to membrane inner leaflet lipids. Homooligomerizes within the membrane and forms pores of 10-15 nanometers (nm) of inner diameter, triggering pyroptosis.
