GSTA3 also known as Glutathione S-transferase alpha 3 is an enzyme involved in the detoxification processes. It exhibits a molecular mass of approximately 25 kDa. GSTA3 functions by catalyzing the conjugation of reduced glutathione to hydrophobic and electrophilic compounds facilitating their solubility and excretion. It is expressed mainly in the liver albeit at various levels in other tissues including kidney and pancreas. This enzyme plays an important role in preventing cellular damage from reactive metabolites and toxins.
Biological function summary

GSTA3 acts in the neutralization of oxidative stress by reducing lipid hydroperoxides to their alcohol forms and supporting the intracellular antioxidant defense. While GSTA3 functions as a monomer it can form homodimers or heterodimers with other isoenzymes enhancing its functional diversity. This capability allows GSTA3 to partake in metabolizing a wide array of endogenous and xenobiotic substrates maintaining cellular health.

Pathways

GSTA3 plays a part in the glutathione metabolism and xenobiotic clearance. It plays an essential role in the detoxification pathway acting in conjunction with other glutathione S-transferases such as GSTM1 and GSTP1. Together these proteins form a network that facilitates detoxification and protects against oxidative damage in tissues. The capacity of GSTA3 to mitigate harmful chemical interactions places it in central biological pathways that uphold cellular integrity.

Alterations or deficiencies in GSTA3 expression or function associate with an increased risk of liver diseases and certain cancers. Due to its role in detoxifying carcinogens compromised GSTA3 activity can lead to carcinogenesis particularly in hepatic tissues. Additionally research reveals interactions with proteins like GSTM1 in the context of cancer where combined effects further influence susceptibility to disease. Understanding the mechanistic role of GSTA3 in these conditions may provide insights into therapeutic interventions and biomarker development.
