The GSTM1 GSTM2 GSTM4 and GSTM5 proteins belong to the glutathione S-transferase family also known as GST mu class. These proteins play a role in detoxification by catalyzing the conjugation of reduced glutathione to a wide number of exogenous and endogenous hydrophobic electrophiles. Each GSTM isoform weighs roughly 25-28 kDa. They are expressed in various tissues most abundantly in the liver kidney and lungs.
Biological function summary

The glutathione S-transferase mu proteins protect cells from oxidative stress. They participate in the detoxification of reactive oxygen species and xenobiotics. These proteins often function as part of the larger enzyme complex that incorporates glutathione. Their activity impacts cellular survival by managing toxic substances that accumulate.

Pathways

GSTM proteins fit into important processes such as the xenobiotic metabolism pathway and the glutathione metabolism pathway. They work closely with other enzyme families like GST pi and GST alpha in the conjugation phase of detoxification. The interaction with proteins like cytochrome P450 responsible for the initial oxidation of toxins is also notable.

Alterations in GSTM proteins associate with conditions like cancer and asthma. For instance GSTM1 null variants often link to an increased risk of lung cancer because of impaired detoxification. Additionally these proteins connect with inflammation-related proteins in asthma influencing disease progression and response to treatment.
