The GSTM1 protein also known as Glutathione S-transferase Mu 1 plays important roles in cellular detoxification processes. It belongs to the glutathione S-transferase family of enzymes which are involved in the conjugation of reduced glutathione to xenobiotic substrates for the purpose of detoxification. GSTM1 has a molecular mass of approximately 26 kDa. This protein expresses mostly in liver and kidney tissues but has presence in other tissues like lung and testis as well.
Biological function summary

GSTM1 acts in metabolism of endogenous and xenobiotic compounds. It does not work alone but rather functions as part of enzyme complexes that alter chemical structure of toxic substances making them safer for cellular systems. GSTM1 helps in protecting cells from damage caused by oxidative stress. The interaction of this protein with glutathione provides a protective mechanism against toxic chemicals such as carcinogens and electrophiles.

Pathways

GSTM1 has significant involvement in metabolic detoxification and the glutathione-mediated pathway. It plays a role in the metabolism of drugs and clearance of harmful compounds in these pathways. It often interacts with other proteins like GSTP1 and GSTT1 within these pathways working together to facilitate cellular defense mechanisms. GSTM1 contributes importantly to maintaining cellular redox homeostasis by its enzymatic activities.

GSTM1 shows a connection to various cancers and cardiovascular diseases. The absence or null genotype of GSTM1 associates with higher risk of developing certain cancers like bladder and lung cancer due to impaired detoxification capacity. In cardiovascular diseases the protein's deficiency can disrupt normal oxidative stress balance. GSTP1 operates alongside GSTM1 in pathological situations with their combined functions influencing disease susceptibility and progression.
