GTF2F2 also known as RAP74 forms a critical component of the general transcription factor IIF. It possesses an approximate mass of 74 kDa. This protein ubiquitously expresses in human cells acting as a cofactor within RNA polymerase II transcription machinery. GTF2F2 binds with RNA polymerase II influencing transcription initiation by stabilizing the transcription pre-initiation complex.
GTF2F2 plays a significant role in enhancing the process of transcription elongation. It joins the multi-protein transcription factor IIF complex collaborating with subunits such as TFIIF. GTF2F2's activity is necessary for promoter clearance and efficient RNA synthesis from DNA templates bridging interactions between transcription factors and the polymerase.
Involvement of GTF2F2 is important in the transcriptional regulation pathway. It coordinates with RNA polymerase II and influences the transcription cycle particularly pathways related to transcription elongation. GTF2F2 shares interactions with other transcription-associated proteins including TFIIB and TFIIH which further integrate into these pathways to optimize transcription efficiency and fidelity.
Studies link GTF2F2 to neurological disorders including mental retardation. The protein's disruption might alter transcriptional regulation leading to developmental anomalies. In cancer it holds a connection with proteins like p53 where changes in transcription factor activity influenced by GTF2F2 can affect cancer progression or response to treatment through disruption of proper gene expression.
