GTPBP4 also known as NOG1 is a highly conserved protein involved in ribosome biogenesis. The protein has a molecular mass of approximately 72 kDa. GTPBP4 is a GTP-binding protein which belongs to the circularly permuted GTPase (cpGTPase) family. It typically locates in the nucleolus where it is involved in ribosomal RNA processing and assembly of the large ribosomal subunit. Expression of GTPBP4 is widespread in human tissues indicating its important role in cellular processes.
Biological function summary

The GTPBP4 protein contributes to the proper formation of the 60S ribosomal subunit through its role in ribosome assembly. It operates alongside other proteins as part of a complex. This role is important for maintaining normal levels of protein synthesis within cells. In particular GTPBP4 interacts with multiple ribosomal and nucleolar proteins to regulate ribosome assembly efficiently. Its activity ensures the structural maturation needed for the large subunit to join the small subunit during translation.

Pathways

GTPBP4 is a part of the ribosome biogenesis pathway. This pathway is important for the production of ribosomal RNA and assembly of ribosomal components. GTPBP4 works in conjunction with proteins like NPM1 and other nucleolar components to maintain the fidelity of ribosome production. It also plays a role in ensuring that the ribosomal subunits are properly assembled and functional for mRNA translation therefore linking the biogenesis of ribosomes with protein synthesis pathways.

Alterations in GTPBP4 expression or function can lead to various diseases such as cancer. GTPBP4 has connections to nucleolar stress responses which affect tumorigenesis. It interacts with proteins like MYC influencing cell proliferation and apoptosis. Dysregulation of GTPBP4 may disrupt ribosome biogenesis leading to imbalances in cell growth and division. It can also be involved in ribosomopathies disorders caused by defects in ribosome production impacting cellular protein synthesis and growth.
