Intracellular Flow Cytometry analysis of HepG2 (Human hepatocellular carcinoma epithelial cell) cells labeling HABP2 with purified ab181837 at 1/20 dilution (10 μg/mL) (Red). Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) secondary antibody was used at 1/2000. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).

The Hyaluronan-Binding Protein 2 (HABP2) also known as Factor VII activating protease is a serine protease with an approximate mass of 65 kDa. This protein gets expressed mainly in the liver and secreted into the bloodstream. HABP2 participates in extracellular matrix interactions by binding to hyaluronan. It plays a role in coagulation processes. The protein also influences angiogenesis highlighting its mechanical involvement in vascular biology.
Biological function summary

HABP2 functions as a regulator in the maintenance of blood homeostasis and tissue remodeling. It takes part in fibrinolysis the process that prevents blood clots from growing and becoming problematic. HABP2 works together with fibrin to facilitate clot breakdown. It may also interact with plasminogen activators within these complexes to accomplish its biological functions effectively.

Pathways

HABP2 integrates into the fibrinolytic and coagulation pathways. It contributes to ensuring that the balance between clot formation and breakdown is maintained. The protein shows interactions with other components like urokinase-type plasminogen activator (uPA) which also plays a role in fibrinolysis. Cleaves the alpha-chain at multiple sites and the beta-chain between 'Lys-53' and 'Lys-54' but not the gamma-chain of fibrinogen and therefore does not initiate the formation of the fibrin clot and does not cause the fibrinolysis directly (PubMed : 11217080). It does not cleave (activate) prothrombin and plasminogen but converts the inactive single chain urinary plasminogen activator (pro-urokinase) to the active two chain form (PubMed : 10754382, PubMed : 11217080). Activates coagulation factor VII (Probable). May function as a tumor suppressor negatively regulating cell proliferation and cell migration (PubMed : 26222560).
