HAPLN1 also known as hyaluronan and proteoglycan link protein 1 is an important structural component in the extracellular matrix. This protein links hyaluronan to proteoglycans which contributes to the integrity and function of the matrix. HAPLN1 has a molecular mass of approximately 39 kDa and is expressed in various tissues including cartilage and nervous tissues. Its expression plays a significant role in maintaining the properties of these tissues.
Hyaluronan and proteoglycan link protein 1 supports the organization and stabilization of the extracellular matrix network. As part of this framework HAPLN1 forms complexes with hyaluronan and aggrecan ensuring structural stability and resilience in tissues experiencing mechanical stress. The association with such complexes impacts cellular signaling and tissue hydration which are important for maintaining tissue homeostasis and function.
HAPLN1 participates in extracellular matrix organization and cell signaling processes. It is actively involved in the aggrecan pathway which influences cartilage development and repair. HAPLN1 is also connected to proteins like aggrecan and link proteins that help mediate interactions within the matrix and regulate cellular responses to mechanical stress.
Abnormal expression or function of HAPLN1 has links to osteoarthritis and intervertebral disc degeneration. In these conditions the disruption of extracellular matrix composition alters tissue integrity and function. An imbalance involving HAPLN1 along with proteins like aggrecan contributes to the pathogenesis and progression of these disorders affecting joint and disc health.

Intracellular Flow Cytometry analysis of MCF-7 (human breast carcinoma) cells labeling HAPLN1 with purified Anti-HAPLN1 antibody [EPR6338] ab181997 at 1/2000 dilution (red). The secondary antibody was Goat anti rabbit IgG (Alexa Fluor® 488) at 1/2000 dilution. A Rabbit monoclonal IgG (Black) was used as the isotype control and cells without incubation with primary antibody and secondary antibody (Blue) were used as unlabeled control.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (Anti-HAPLN1 antibody [EPR6338] ab181997).
