Supplementary information

Haspin also known as Germ Cell-Specific Gene 2 (GSG2) is a serine/threonine-protein kinase with a molecular weight of approximately 86 kDa. Haspin expresses predominantly in the testes but researchers have detected it in other tissues too. This kinase is well recognized for its role during cell division especially in the regulation of chromosome alignment and segregation by phosphorylating histone H3 at threonine 3.
Biological function summary

Haspin kinase plays a significant role in mitosis and meiosis. It orchestrates the proper attachment of microtubules to kinetochores by modifying chromosomal components. Haspin is part of a larger protein complex during cell division working closely with the chromosomal passenger complex to ensure accurate segregation of chromosomes. Its activity ensures the integrity and stability of cellular division processes.

Pathways

Haspin kinase functions centrally in cell cycle regulation and mitotic spindle assembly pathways. It interacts with key proteins in these pathways such as Aurora B kinase. The phosphorylation of histone H3 by haspin establishes a critical docking site for proteins in the chromosomal passenger complex facilitating proper chromosome alignment and spindle checkpoint activation.

Incorrect regulation of haspin activity associates with various cancers and chromosomal instability disorders. Overexpression or mutations in haspin can contribute to tumorigenesis by disrupting normal cell division. Additionally haspin's interaction with the chromosomal passenger complex proteins like survivin influences the development of colorectal cancer.

Target data

Serine/threonine-protein kinase that phosphorylates histone H3 at 'Thr-3' (H3T3ph) during mitosis. May act through H3T3ph to both position and modulate activation of AURKB and other components of the chromosomal passenger complex (CPC) at centromeres to ensure proper chromatid cohesion, metaphase alignment and normal progression through the cell cycle.
