Supplementary information

The HBE protein also known as hepatitis B virus e antigen (HBeAg) displays significant mechanical functions in the viral life cycle. It has a mass of approximately 17–25 kDa and expresses mainly in the cells of the liver during hepatitis B virus replication. HBE gets produced during the conversion of the precore protein into its mature form which allows it to function effectively as a viral antigen. 'Anti-HBE' and 'anti-HBeAg' antibodies target this antigen. The protein plays a role in immune tolerance mechanisms by modulating host immune responses against the hepatitis B virus.
Biological function summary

The HBE protein contributes to the persistence of hepatitis B virus infection. It's part of the viral replication complex and helps maintain viral chronicity by repressing immune activation. The presence of HBE in the host leads to a reduction in the recognition of infected hepatocytes by the immune system allowing the virus to persist longer. Identifying the presence of 'HBE antigen' and 'anti-HBE' antibodies helps monitor the progression of hepatitis B infection. This antigen is a marker for active viral replication and infectiousness.

Pathways

HBE integrates into the hepatitis B virus life cycle and immune response pathways. It involves the regulation of host immune responses particularly the adaptive immune system's recognition and clearance of infected hepatocytes. HBeAg influences the pathways by interacting with proteins related to immune checkpoints ensuring viral persistence. These interactions alter the balance of pro-inflammatory and anti-inflammatory signals affecting the body's ability to mount an effective immune response against the infected liver cells.

HBE plays an important role in chronic hepatitis B infection and liver disease progression. It associates with reduced inflammation in chronic hepatitis B patients often correlating with lower liver damage. However persistent expression of HBE can contribute to the development of hepatitis B-related liver cirrhosis due to prolonged immune suppression. The production of 'anti-HBE' and 'anti-HBeAg' antibodies against HBE indicates the host's immune status and helps in determining the phase of infection. Understanding the interactions between HBE and other hepatic proteins assists clinicians in making decisions regarding hepatitis B treatment strategies.
