Immunocytochemistry/ Immunofluorescence - Anti-HCoV-229E Nucleocapsid protein antibody [abd210] (AB316285)
Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized 293T (human embryonic kidney epithelial cell) transfected with a HCoV-229E Nucleocapsid protein expression vector containing a myc tag. cells labelling HCoV-229E Nucleocapsid protein with ab316285 at 1/200 (4.985 ug/ml) dilution, followed by Goat anti-Human IgG (H+L) Cross-Adsorbed Secondary Antibody (Alexa Fluor® 488) antibody at 1/1000 (2ug/ml) dilution (Green).
Confocal image showing cytoplasmic staining in 293T cells (shown in green) transfected with HCoV-229E Nucleocapsid protein expression vector containing a myc-His-tag®. The counterstain was observed in red. Nuclear DNA was labeled with DAPI (shown in blue).
Image was taken with a confocal microscope(Leica-Microsystems, TCS SP8).
ab223895 Anti-Myc tag Mouse monoclonal antibody (Alexa Fluor® 647) was used to counterstain at 1/100 (5ug/ml) dilution (Red). The Nuclear counterstain was DAPI (Blue).
Secondary antibody only control : Secondary antibody is Goat anti-Human IgG (H+L) Cross-Adsorbed Secondary Antibody (Alexa Fluor® 488) at 1/1000 (2ug/ml) dilution.

Biological function summary
The N protein directly impacts viral replication and transcription. It forms part of a larger ribonucleoprotein complex that organizes the viral genome into a functional and protected structure. This complex interacts with other viral proteins to facilitate the replication process. The N protein also boosts the efficiency of the viral RdRp (RNA-dependent RNA polymerase) in transcribing the RNA genome ensuring that viral replication proceeds efficiently and effectively within host cells.
Pathways
The N protein integrates into the coronavirus replication-transcription complex. It coordinates with the viral RdRp and other non-structural proteins of the virus to ensure a streamlined replication process. This protein interacts with cellular pathways that include stress response and immune pathways potentially modulating host immune responses to benefit viral survival and replication. These interactions with host cell pathways may influence the efficiency of viral replication and the pathogenicity of the virus.
