Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. Involved in muscle maturation via its interaction with the myocyte enhancer factors such as MEF2A, MEF2C and MEF2D. Involved in the MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer. Deacetylates HSPA1A and HSPA1B at 'Lys-77' leading to their preferential binding to co-chaperone STUB1 (PubMed:27708256).

HDAC4 HDAC5 and HDAC9 are types of histone deacetylases abbreviated as HDACs. These enzymes remove acetyl groups from histone and non-histone proteins affecting gene expression by changing chromatin structure. HDAC4 has a molecular weight of about 140 kDa. HDAC4 and HDAC5 are highly expressed in the brain heart and skeletal muscle while HDAC9 is more abundant in the heart and skeletal muscle. They act as transcriptional repressors by interacting with various corepressor complexes.
Histone deacetylases 4 5 and 9 influence gene expression and cellular differentiation. These proteins often form part of larger corepressor complexes such as the N-CoR and SMRT complexes allowing them to modulate transcriptional activity. They play significant roles in muscle differentiation and neural development interacting with transcription factors like MEF2 to regulate various genes involved in these processes.
Histone deacetylases like HDAC4 5 and 9 are critical components of signaling pathways related to calcium/calmodulin-dependent calcineurin signal transduction. This pathway influences muscle development and function where they interact with MEF2 family transcription factors to control gene expression. Additionally they are involved in the regulation of the circadian rhythm through their role in the molecular clockwork partnering with proteins like CLOCK and BMAL1.
Alterations in HDAC4 5 and 9 levels or activity have connections to cardiovascular diseases and certain cancers. Dysfunctional HDAC activity can lead to hypertrophy in cardiac cells affecting heart function. Cancer progression often involves aberrant histone deacetylation with HDACs modulating oncogenic transcription factors. These HDACs closely interact with cancer-related proteins like MYC and p53 influencing tumor suppression or progression depending on cellular context.
