HECTD3 was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab241413 at 6 μg/reaction. Western blot was performed from the immunoprecipitate using ab241413 at 1 μg/ml.

HECTD3 also known as hectd3 ubiquitin protein ligase acts as an E3 ubiquitin ligase. It plays an important role in tagging proteins for degradation through the ubiquitin-proteasome system. The molecular weight of HECTD3 is approximately 110 kDa. HECTD3 is mainly expressed in the brain and various immune cells indicating its involvement in neurological and immune functions.
Biological function summary

The enzyme facilitates the regulation of proteins by adding ubiquitin molecules which marks them for degradation or signaling. This function helps control the levels of target proteins and maintain cellular homeostasis. HECTD3 often forms part of a protein complex enabling its ligase activity to be moderated by interacting proteins.

Pathways

The enzyme integrates into the ubiquitin-proteasome pathway and the NF-kB signaling pathway. In the ubiquitin-proteasome system HECTD3 collaborates with other E2 and E1 enzymes to achieve efficient ubiquitination. It interacts directly with IκB proteins in the NF-kB signaling pathway impacting inflammatory and immune responses by controlling the stability and activity of these proteins.

HECTD3's malfunctions correlate to various cancers and immune disorders. For example its dysregulation affects certain types of leukemia where it modifies the degradation process of tumor suppressor proteins. Moreover HECTD3 interacts with proteins such as TP53 which also plays a significant role in the development of cancers. Changes in HECTD3 expression in immune cells influence immune disorders connecting it to autoimmune diseases where immune regulation becomes disrupted.

E3 ubiquitin ligases accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfers the ubiquitin to targeted substrates. Mediates ubiquitination of TRIOBP and its subsequent proteasomal degradation, thus facilitating cell cycle progression by regulating the turn-over of TRIOBP. Mediates also ubiquitination of STX8 (By similarity).
