HERV-FRD also known as ERVFRD-1 or Syncytin-2 is a protein encoded by the human endogenous retrovirus family F envelope (env) gene. This protein has a molecular mass of approximately 63 kDa. HERV-FRD is strongly expressed in the placenta particularly in the syncytiotrophoblast layer which plays an important role during pregnancy. It functions as an envelope glycoprotein and contributes to cellular fusion processes essential for the formation of syncytiotrophoblasts.

Biological function summary

HERV-FRD plays an important role in forming syncytiotrophoblasts which are vital for the function of the human placenta. This protein is a component of a larger protein complex that facilitates membrane fusion a critical step in placental development. The fusion process mediated by HERV-FRD is necessary for proper nutrient and gas exchange between the mother and the developing fetus highlighting its role in supporting fetal development.

Pathways

HERV-FRD interacts with the Syncytin-mediated cell fusion pathway which is important for placental morphogenesis. This protein also interacts with other syncytin proteins like Syncytin-1 which share similar roles in promoting trophoblast fusion. Both proteins derived from different human endogenous retroviruses are integral components of mechanisms that ensure proper placental structure and function.

Abnormalities in HERV-FRD function have been associated with complications such as preeclampsia and certain reproductive failures. Altered expression or function of this protein may affect placental development potentially impacting maternal and fetal health. Additionally HERV-FRD's role in cellular fusion links it indirectly to disorders involving aberrant cell fusion events often seen in certain gestational trophoblastic diseases alongside related proteins such as Syncytin-1.

This endogenous retroviral envelope protein has retained its original fusogenic properties and participates in trophoblast fusion and the formation of a syncytium during placenta morphogenesis. The interaction with MFSD2A is apparently important for this process (PubMed : 18988732).. Endogenous envelope proteins may have kept, lost or modified their original function during evolution but this one can still make pseudotypes with MLV, HIV-1 or SIV-1 virions and confer infectivity. Retroviral envelope proteins mediate receptor recognition and membrane fusion during early infection. The surface protein mediates receptor recognition, while the transmembrane protein anchors the envelope heterodimer to the viral membrane through one transmembrane domain. The other hydrophobic domain, called fusion peptide, mediates fusion of the viral membrane with the target cell membrane (PubMed : 14694139).
