HES6 also known as Hairy and Enhancer of Split-6 is a protein involved in transcriptional regulation. It has a molecular mass of approximately 25 kilodaltons. Researchers find HES6 expression in multiple tissues including the brain muscle and pancreas. As a member of the bHLH (basic helix-loop-helix) transcription factor family HES6 negatively regulates transcription typically by antagonizing the activity of other transcription factors.
Biological function summary

HES6 influences cell differentiation and the timing of neurogenesis. It does not commonly act as part of a large complex but rather modulates activity by interacting directly with other transcription factors and regulatory proteins. Through these interactions HES6 helps control gene expression patterns important for the development of neural and muscular structures.

Does not bind DNA itself but suppresses both HES1-mediated N box-dependent transcriptional repression and binding of HES1 to E box sequences. Also suppresses HES1-mediated inhibition of the heterodimer formed by ASCL1/MASH1 and TCF3/E47, allowing ASCL1 and TCF3 to up-regulate transcription in its presence. Promotes cell differentiation (By similarity).
