Required for the normal development of the forebrain, eyes and other anterior structures such as the olfactory placodes and pituitary gland. Possible transcriptional repressor. Binds to the palindromic PIII sequence, 5'-AGCTTGAGTCTAATTGAATTAACTGTAC-3'. HESX1 and PROP1 bind as heterodimers on this palindromic site, and, in vitro, HESX1 can antagonize PROP1 activation.

HESX1 also known as Homeobox expressed in ES cells 1 is a homeobox gene encoding a protein with a molecular mass of approximately 22 kDa. This protein acts as a transcriptional repressor and is expressed in the early stages of embryonic development particularly within the anterior pituitary gland and developing forebrain. The HESX1 protein interacts with DNA sequences to regulate the expression of target genes essential for forebrain and pituitary formation.
HESX1 functions as a transcriptional repressor influencing the development of head and pituitary embryogenesis. This protein is not part of a larger complex but works by binding directly to promoter regions of DNA affecting transcriptional activity. HESX1 plays a critical role in regulating the expression of genes necessary for early development and cellular differentiation ensuring proper formation of the forebrain and pituitary gland.
HESX1 is involved in important developmental pathways including the Wnt signaling and Sonic Hedgehog pathways. These pathways govern cellular processes such as proliferation differentiation and morphogenesis. HESX1's role in these pathways connects it with proteins like β-catenin in the Wnt signaling and SHH protein in the Sonic Hedgehog pathway which are essential for maintaining the balance of cell growth and organ development.
Mutations in HESX1 are associated with septo-optic dysplasia and combined pituitary hormone deficiency. These conditions affect brain and pituitary development leading to clinical manifestations like optic nerve hypoplasia and hormone deficiencies.
