Supplementary information

HEXIM2 also known as Hexamethylene Bisacetamide-Inducible Protein 2 is a protein with a molecular mass of approximately 36 kDa. It functions as a regulatory protein with roles in transcription modulation. HEXIM2 is expressed in various human tissues with notable expression levels in the heart and skeletal muscle. This protein acts as a part of the regulation network that interacts with RNA polymerase II during the transcription process.
Biological function summary

HEXIM2 plays a role in the inhibitory regulation of transcription. It forms part of the 7SK small nuclear ribonucleoprotein (snRNP) complex which is known for controlling the activity of the positive transcription elongation factor b (P-TEFb). This complex regulates RNA polymerase II by inhibiting its transcriptional elongation capability therefore controlling gene expression. The presence of HEXIM2 in this complex indicates its involvement in modulating gene expression through RNA interference pathways.

Pathways

HEXIM2 operates within the transcription elongation regulation pathway and the cell cycle control pathway. In the transcription elongation regulation pathway it contributes significantly to the inhibition of P-TEFb which associates with Cyclin T1 and CDK9. These proteins are vital for allowing RNA polymerase II to proceed with transcription. HEXIM2's involvement ensures the proper timing and modulation of transcriptional activities controlling cellular functions and responses.

HEXIM2 has associations with cancer and cardiac-related conditions. Its role in transcription regulation links it to various forms of cancer where aberrant gene expression is a hallmark. Additionally HEXIM2's expression in cardiac tissues suggests implications in cardiac disorders. Interactions with proteins like Cyclin T1 and CDK9 which are also involved in these conditions highlight potential pathways where HEXIM2 might contribute to disease progression or resistance mechanisms.
