HGFA Inhibitor 2 also known as HAI-2 is a serine protease inhibitor involved in regulating the activity of hepatocyte growth factor activator (HGFA). The approximate molecular mass of HAI-2 is 54 kDa. This inhibitor is widely expressed in many tissues but shows especially high expression in the epithelial tissues including the liver kidney and gastrointestinal tract. It often acts near sites of tissue injury or regeneration due to its role in controlling proteolytic activity.
Biological function summary

HGFA Inhibitor 2 is involved in the control of protease-mediated signaling pathways by functioning as a member of a protease inhibitor complex. It plays a role in modulating the activity of proteolytic enzymes which influence tissue remodeling cell growth and angiogenesis. By inhibiting HGFA HAI-2 regulates the conversion of inactive hepatocyte growth factor (HGF) into its active form therefore controlling processes that are important for epithelial cell dynamics.

Pathways

HGFA Inhibitor 2 contributes to the HGF/c-Met signaling pathway and impacts the plasminogen activation system. Through the regulation of HGFA HAI-2 inadvertently influences the activation of c-Met a receptor tyrosine kinase. This interaction is key for numerous processes including cell proliferation motility and morphogenesis. Additionally by affecting the plasminogen activation cascade it helps to modulate extracellular matrix degradation and cellular migration through its association with proteins like uPA and plasminogen.

HGFA Inhibitor 2 relates to conditions such as cancer and chronic kidney disease. It is significant in cancer biology due to its influence on tumor growth and metastasis given its role in the activation of the HGF/c-Met pathway frequently implicated in various cancers. In chronic kidney disease the dysregulation of HAI-2 can disrupt normal protease activity affecting kidney function and potentially contributing to disease progression. Related proteins in these contexts include HGFA and c-Met which are involved in the alteration of cellular behavior in these pathologies.
