HHLA3 also known as Human Endogenous Retrovirus (HERV)-H Long Terminal Repeat-Associating protein 3 is a cell surface protein with a predicted mass of approximately 32 kDa. This protein is a member of the B7 family which plays a role in immune regulation. HHLA3 is expressed in a range of tissue types with high levels in the placenta and moderate levels in peripheral blood mononuclear cells (PBMCs). Its expression pattern indicates potential involvement in immune system processes.
Biological function summary

HHLA3 functions in immune modulation and may inhibit T-cell responses. Researchers have yet to confirm its participation in protein complexes but interactions with other B7 family proteins suggest a potential role. These interactions possibly regulate immune checkpoints which control immune response strength and are important for preventing autoimmunity and maintaining self-tolerance.

Pathways

Researchers find HHLA3 relevant to the immune checkpoint pathways and T-cell receptor signaling. Through these pathways HHLA3 might influence the activation and inhibition of T-cells.
