Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized HeLa (human cervical adenocarcinoma epithelial cell) and HeLa treated with CoCl2 (0.5 mM) for 6 hr cells labelling HIF-1 alpha with ab325355 at 1/50 dilution, followed by ab150157 Goat Anti-Rat IgG H&L (Alexa Fluor® 488) antibody at 1/1000 dilution (Green).

Blocking and diluting buffer and concentration : 5% NFDM/TBST.

In Western blot, ab325355 was shown to bind specifically to HIF-1 alpha. A band was observed at 110 kDa in wild-type HCT 116 cell lysates with whereas no signal observed at this size in HIF-1 alpha knockout cell line ab255394.

Lysates were freshly made and used for Western blotting immediately to minimize protein degradation.

In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (ab181602) (1 : 200000) (36KDa).

Target data

Functions as a master transcriptional regulator of the adaptive response to hypoxia (PubMed : 15225651, PubMed : 17981124, PubMed : 22009797). Under hypoxic conditions, activates the transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, HILPDA, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia (PubMed : 15225651, PubMed : 17981124, PubMed : 22009797). Plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease (PubMed : 22009797). Heterodimerizes with ARNT; heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters (PubMed : 26245371). Activation requires recruitment of transcriptional coactivators such as CREBBP and EP300. Activity is enhanced by interaction with NCOA1 and/or NCOA2. Interaction with redox regulatory protein APEX1 seems to activate CTAD and potentiates activation by NCOA1 and CREBBP. Involved in the axonal distribution and transport of mitochondria in neurons during hypoxia (By similarity).
