HIP1R also known as Huntingtin-interacting protein 1 related is an endocytic adaptor protein. This protein weighs approximately 116 kDa. It primarily facilitates clathrin-mediated endocytosis by linking endocytic vesicles to the actin cytoskeleton. HIP1R is enriched in neuronal tissues particularly within the brain and is present in other tissues like muscle and liver. The protein contains a domain structure having an ENTH domain and coiled-coil regions which allow it to interact with other cellular components.
Biological function summary

HIP1R contributes to membrane trafficking and actin dynamics. As part of the endocytic machinery HIP1R directly interacts with clathrin and actin fostering vesicle motility and shaping membrane curvature. The protein resides within a larger protein complex that includes clathrin actin and other endocytic proteins. Its role in vesicle formation and transport is essential for maintaining cellular homeostasis by controlling receptor internalization and sorting.

Pathways

HIP1R participates in the clathrin-mediated endocytosis and actin cytoskeleton regulation pathways. It shares functional connections with proteins such as clathrin and actin emphasizing its role in vesicular dynamics and membrane trafficking. The protein ensures effective communication and transport within cells by coordinating with these pathways. These processes contribute to essential cellular functions like nutrient uptake and signal transduction.

HIP1R associates with neurodegenerative conditions and cancer. Altered expression of HIP1R can occur in pathological states impacting endocytic and cytoskeletal functions. Its interaction with Huntingtin another significant protein connects it to Huntington’s disease where proper endocytic action becomes disrupted. In cancer dysregulation of endocytosis-related proteins including HIP1R leads to aberrant cell signaling and proliferation linking HIP1R to oncogenic pathways.
