ab241151 was tested in Peptide Array against 501 different modified and unmodified histone peptides; each peptide is printed on the array at six concentrations (each in triplicate). The concentration of ab241151 was 1/2500 dilution, the secondary used was Goat Anti-Rabbit IgG, (H+L), Fluo 647nm conjugated at 1/50000 dilution. Circle area represents affinity between the antibody and a peptide : all antigen-containing peptides are displayed as red circles, all other peptides as blue circles. The affinity is calculated as the area under the curve when antibody binding values are plotted against the corresponding peptide concentration. Each circle area is normalized to the peptide with the strongest affinity. This blot was produced using a 4-20% Criterion TGX gel in 1X SDS buffer. The gel was run at 180V for 40 minutes before being transferred onto a Nitrocellulose membrane at 25V for 7 minutes using the Trans-Blot turbo transfer system. The membrane was then blocked for an hour using 3% milk before being incubated with ab241151 overnight at 4°C. Antibody binding was detected using a goat anti-Rabbit antibody conjugated to HRP, and visualised using Bio-Rad Clarity Western ECL substrate

All batches of ab241151 are tested in Peptide Array against peptides to different Histone H2, H3 and H4 modifications. Six dilutions of each peptide are printed on to the Peptide Array in triplicate and results are averaged before being plotted on to a graph. The protein expressed by gene H3C1 is a core component of the nucleosome, which is essential for wrapping and compacting DNA into chromatin. This process restricts DNA accessibility to cellular machineries that use DNA as a template. Consequently, histones are central to transcription regulation, DNA repair, DNA replication, and chromosomal stability.
