Immunocytochemistry/ Immunofluorescence - Anti-Histone H3 (phospho T3, di methyl K4) antibody (AB272139)

Immunofluorescence analysis of HeLa cells labeling Histone H3 (green) using ab272139 at 1 : 50, followed by Dylight 488 secondary antibody at 1 : 10,000. Localization : Histone H3 [p Thr3, Monomethyl Lys4] is nuclear and chromosomal. Countersatin : nuclei and aplpha-tubulin were coexpressed with DAPI (blue) and Dylight 550 (red).

ChIP - Anti-Histone H3 (phospho T3, di methyl K4) antibody (AB272139)

Chromatin Immunoprecipitation of ab272139. Chromatin from one million formaldehyde cross-linked HeLa cells was used with 2ug of ab272139 and 20ul of magnetic IgG beads per immunoprecipitation. A no antibody (No Ab) control was also used. Immunoprecipitated DNA was quantified using quantitative real-time PCR and SYBR green dye, then normalized to the non-precipitated input chromatin, which is equal to one.

Specificity

This antibody reacts with human Histone H3.2. A BLAST analysis was used to suggest cross-reactivity with Human, mouse, and C. elegans. Predicted to react with many species including rat, chicken, Xenopus, Drosophila, and plant based on 100% sequence homology. Cross-reactivity with Histone H3 from other sources has not been determined.

Target data

The protein expressed by gene H3C1 is a core component of the nucleosome, which is essential for wrapping and compacting DNA into chromatin. This process restricts DNA accessibility to cellular machineries that use DNA as a template. Consequently, histones are central to transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated through a complex set of post-translational modifications of histones, known as the histone code, and through nucleosome remodeling. This supplementary information is collated from multiple sources and compiled automatically.
