Function

Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

  • ChIP - Anti-Histone H3 (tri methyl K64) antibody (ab195483), expandable thumbnail

    ChIP - Anti-Histone H3 (tri methyl K64) antibody (ab195483)

ChIP analysis using K562 cells, ab195483 and optimized PCR primer sets for qPCR. ChIP was performed using sheared chromatin from 1 million cells. A titration of the antibody consisting of 1, 2, 5, and 10 μg per ChIP experiment was analysed. IgG (2 μg/IP) was used as negative IP control. QPCR was performed with primers for the promoter of the active GAPDH and EIF4A2 genes, used as negative controls, and for the Sat2 and Sata satellite repeats, used as positive controls.

  • Dot Blot - Anti-Histone H3 (tri methyl K64) antibody (ab195483), expandable thumbnail

    Dot Blot - Anti-Histone H3 (tri methyl K64) antibody (ab195483)

Dot Blot analysis was performed to test the cross reactivity of ab195483 against Histone H3 (tri methyl K64) (H3K64me3) with peptides containing different modifications of histone H3 and H4 or the unmodified H3K64 sequence. One hundred to 0.2 pmol of peptide containing the respective histone modification were spotted on a membrane. ab195483 was used at 1/5,000 dilution.

  • Western blot - Anti-Histone H3 (tri methyl K64) antibody (ab195483), expandable thumbnail

    Western blot - Anti-Histone H3 (tri methyl K64) antibody (ab195483)

The antibody was diluted 1/100 in TBS-Tween containing 5% skimmed milk.

All lanes: Western blot - Anti-Histone H3 (tri methyl K64) antibody (ab195483) at 1/100 dilution

All lanes: histone extracts from HeLa cells at 30 µg

    Predicted band size: 15 kDa
