HKDC1 was immunoprecipitated from 0.35 mg HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate with ab209846 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab209846 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.

Lane 1 : HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate 10 μg (Input).

Lane 2 : ab209846 IP in HeLa whole cell lysate (+).

Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab209846 in HeLa whole cell lysate (-).

Blocking and dilution buffer and concentration : 5% NFDM/TBST.

Exposure time : 3 seconds.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab209846).

The HKDC1 protein also known as Hexokinase Domain Containing 1 participates in glucose metabolism by phosphorylating glucose to produce glucose-6-phosphate. It has a molecular mass of approximately 102 kDa. This protein is ubiquitously expressed with higher levels in the liver pancreas and certain fetal tissues. HKDC1 plays an important role in maintaining glucose homeostasis particularly under conditions that require enhanced glucose utilization.
Biological function summary

Hexokinase Domain Containing 1 functions as part of the hexokinase enzyme family contributing to the first step of glycolysis. It might not form a complex but shares similar activity with other hexokinase family members supporting cellular energy supply. The presence of HKDC1 in various tissues suggests its involvement in both normal and stress-related metabolic regulation adapting the cell’s energy production to different physiological demands.

Pathways

The activity of HKDC1 is integrated into the glycolytic pathway and indirectly affects the insulin signaling pathway. In glycolysis it links to glucokinase and other hexokinase enzymes playing a part in glucose metabolism and energy generation within the cell. Its activity impacts the homeostatic control of glucose concentration suggesting a regulatory role in maintaining the balance between energy input and output in metabolic pathways.

Mutations or altered expression of HKDC1 could be linked to metabolic syndromes such as diabetes and gestational diabetes mellitus. Aberrant HKDC1 function may disturb glucose homeostasis leading to these conditions. Moreover connection with other proteins like GCK which also participates in glucose phosphorylation highlights its potential importance in metabolic diseases indicating that HKDC1 might be a target for therapeutic interventions.

Catalyzes the phosphorylation of hexose to hexose 6-phosphate, although at very low level compared to other hexokinases (PubMed : 30517626). Has low glucose phosphorylating activity compared to other hexokinases (PubMed : 30517626). Involved in glucose homeostasis and hepatic lipid accumulation. Required to maintain whole-body glucose homeostasis during pregnancy; however additional evidences are required to confirm this role (By similarity).
