Immunohistochemistry analysis in neuroblast nuclei from Drosophila stage 15 embryos. The cells were fixed in 4% formaldehyde for 25 minutes, permeabilized with 0.1% Triton X-100/PBS, and blocked in 0.1% Triton X-100/PBS, 2.5% Normal Goat Serum, 2.5% Normal Donkey Serum. The Prospero antibody (ab201326) was at 3.4 μg/mL, overnight at 4°C. The secondary antibody used was Alexa488 conjugated goat anti-rat at 1/400.

Homeobox protein prospero also known as Prox1 is a transcription factor involved in regulating gene expression. Common alternate names for this protein include mR1a and ah1-1. It consists of around 83 kDa in its full-length form. The protein is present in various tissues most notably in the nervous system retina and lymphatic endothelium. In these locations homeobox protein prospero influences cell differentiation and growth by modulating the activity of specific target genes.
