Immunocytochemistry/ Immunofluorescence - Anti-Homeobox protein prospero antibody [P3D4-1F10] (AB201326)
Immunohistochemistry analysis in neuroblast nuclei from Drosophila stage 15 embryos. The cells were fixed in 4% formaldehyde for 25 minutes, permeabilized with 0.1% Triton X-100/PBS, and blocked in 0.1% Triton X-100/PBS, 2.5% Normal Goat Serum, 2.5% Normal Donkey Serum. The Prospero antibody (ab201326) was at 3.4 μg/mL, overnight at 4°C. The secondary antibody used was Alexa488 conjugated goat anti-rat at 1/400.

Homeobox protein prospero plays a significant role in neural development influencing the fate and proliferation of neural stem cells. It acts within the nucleus where it binds consensus DNA sequences to control gene transcription. This protein does not usually operate as part of a large complex instead it often interacts directly with chromatin to exert its effects. Its activity is essential for normal neurogenesis and the formation of functional neural circuits.
Pathways
Homeobox protein prospero is essential for the Hedgehog signaling pathway which is critical in the regulation of stem cell maintenance and differentiation. It works closely with other transcription factors such as Pax6 to ensure proper development of the nervous system. Additionally it is involved in the Wnt signaling pathway where it synergizes with other factors to control cell proliferation and fate determination in developing tissues.
