Paraffin-embedded human colon cancer tissue stained for HOOK3 with ab237529 at a 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

HOOK3 also known as Hook homolog 3 is a protein with a mass of approximately 90 kDa. This protein functions mechanically as a microtubule-binding protein which plays a role in cytoskeleton organization. In terms of cellular location researchers have identified its expression in various tissues with high presence noted in brain tissue. HOOK3 engages in hetero-oligomeric interactions and contributes to the stabilization of organelles within the cell.

Acts as an adapter protein linking the dynein motor complex to various cargos and converts dynein from a non-processive to a highly processive motor in the presence of dynactin. Facilitates the interaction between dynein and dynactin and activates dynein processivity (the ability to move along a microtubule for a long distance without falling off the track). Predominantly recruits 2 dyneins, which increases both the force and speed of the microtubule motor (PubMed : 25035494, PubMed : 33734450). Component of the FTS/Hook/FHIP complex (FHF complex). The FHF complex may function to promote vesicle trafficking and/or fusion via the homotypic vesicular protein sorting complex (the HOPS complex). May regulate clearance of endocytosed receptors such as MSR1. Participates in defining the architecture and localization of the Golgi complex.
