Supplementary information

HOXA1 also known as homeobox protein Hox-A1 is a transcription factor composed of 36 kDa. It contains a conserved DNA-binding domain known as the homeodomain. HOXA1 is encoded by the HOXA1 gene part of the Hox gene family responsible for regulating gene expression during early embryonic development. It gets expressed in specific regions during various developmental stages particularly contributing to the formation of the hindbrain and spinal cord. HOXA1 plays a significant role in controlling morphogenesis and differentiation processes.
Biological function summary

The transcription factor regulates critical developmental processes by influencing the expression of genes involved in anterior-posterior patterning. HOXA1 functions within a complex involving other HOX proteins ensuring precise spatial and temporal gene regulation during development. This regulation facilitates the establishment of distinct body structures and organ systems as the embryo progresses. HOXA1 participates in defining boundaries and segmental structures in embryonic tissues.

Pathways

HOXA1 impacts the retinoic acid signaling pathway and the Notch signaling pathway. It plays a significant role in the retinoic acid signaling axis influencing cell differentiation by modulating retinoid-responsive elements. In the Notch signaling pathway HOXA1 interacts with genes that control cell fate decisions often working in concert with other HOX proteins like HOXB1 and HOXD1. This involves activating downstream effectors vital for embryonic patterning and neurogenesis.

Researchers have linked HOXA1 dysregulation to certain congenital defects and cancers. It associates with congenital conditions such as Bosley-Salih-Alorainy syndrome characterized by abnormalities in facial and cranial nerves alongside problems with hearing and cardiovascular development. In cancer aberrant HOXA1 expression has been observed linking to breast cancer progression hinting at its involvement in tumorigenesis. These connections often involve interactions with proteins such as PAX1 in congenital abnormalities and ERBB2 in cancer highlighting the need for therapeutic explorations targeting these pathways.

Target data

Sequence-specific transcription factor (By similarity). Regulates multiple developmental processes including brainstem, inner and outer ear, abducens nerve and cardiovascular development and morphogenesis as well as cognition and behavior (PubMed : 16155570). Also part of a developmental regulatory system that provides cells with specific positional identities on the anterior-posterior axis. Acts on the anterior body structures. Seems to act in the maintenance and/or generation of hindbrain segments (By similarity). Activates transcription in the presence of PBX1A and PKNOX1 (By similarity).
