IHC image of ALDH5A1/SSADH staining in a section of formalin-fixed paraffin-embedded normal human liver*. Performed on a Leica BOND™ system. The section was pre-treated using heat mediated antigen retrieval with EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20mins. The section was then incubated with ab314408 at 5 µg/ml, for 15 mins at room temperature and detected using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset secondary-only control image is taken from an identical assay without primary antibody. For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times. *Tissue obtained from the Human Research Tissue Bank, supported by the NIHR Cambridge Biomedical Research Centre.

Western blot : HRP Anti-ALDH5A1/SSADH antibody [EPR7794] (ab314408) staining at 1/1000 dilution, shown in black; Mouse anti-CANX [CANX/1543] (ab238078) loading control staining at 1/20000 dilution, shown in red. In Western blot, ab314408 was shown to bind specifically to ALDH5A1. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 5 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibody Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution for 1 h at room temperature, washed again four times before development with a high-sensitivity ECL substrate kit and imaged with 8 minutes exposure time.

ALDH5A1 also known as succinate semialdehyde dehydrogenase (SSADH) functions in the cellular metabolism where it converts succinate semialdehyde to succinate in the presence of NAD+. This enzyme has a molecular weight of approximately 56 kDa and is largely found in the brain as well as in other organs like liver and kidney. ALDH5A1 operates in mitochondria emphasizing its role in cellular respiration and energy production.
Biological function summary

The enzyme ALDH5A1 plays an important role in the breakdown of gamma-aminobutyric acid (GABA) an important neurotransmitter. It is known to be part of the GABA degradation complex which controls the levels of GABA in the central nervous system. Balance in GABA concentration is necessary for proper neurotransmission and neurophysiological functions. Consequently ALDH5A1 helps in maintaining the inhibitory effects GABA has on neurons.

Pathways

ALDH5A1 is significant in the GABA shunt pathway an alternative mechanism for glutamate and GABA metabolism. The enzyme collaborates closely with GABA transaminase which converts GABA into succinate semialdehyde before ALDH5A1 further processes it into succinate. This pathway supports energy production and the regulation of carbon flow through central metabolism.

ALDH5A1 deficiencies link to disorders like Succinic Semialdehyde Dehydrogenase deficiency (SSADH deficiency) and GABA-transaminase deficiency. These conditions result in the accumulation of gamma-hydroxybutyric acid leading to neurological complications. This enzyme interacts with proteins like GABA transaminase which shows the importance of its proper function in preventing neurologic disease.
