Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - HRP Anti-GNAI2 antibody [EPR9469] (AB205782)
IHC image of GNAI2 staining in a section of formalin-fixed paraffin-embedded normal human kidney tissue*, performed on a Leica BONDTM. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with ab205782, 1/250 dilution, for 15 mins at room temperature. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Western blot - HRP Anti-GNAI2 antibody [EPR9469] (AB205782)
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab205782 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.

Brain (Human) Tissue Lysate - fetal normal tissue at 10 µg

Biological function summary
GNAI2 functions as part of heterotrimeric G protein complexes. It couples with receptors on the cell surface transmitting signals from activated receptors to downstream effectors. GNAI2 binds with GDP in its inactive state and exchanges it for GTP upon receptor activation which causes the dissociation of the G protein into G alpha and G beta-gamma subunits continuing the signaling cascade. GNAI2 contributes to the modulation of processes such as neurotransmission immune responses and cell proliferation.
Pathways
GNAI2 plays an important role in the MAPK/ERK pathway and the PI3K/Akt signaling pathway. It interacts with various receptors and proteins to regulate cellular responses to external stimuli. For instance through the MAPK/ERK pathway GNAI2 influences gene expression and cell division while in the PI3K/Akt pathway it plays a role in cell survival and apoptosis. Its interaction with proteins like G-beta and G-gamma subunits emphasizes its integrative function in coordinating signal transduction.
