Glutathione S-transferase that catalyzes the nucleophilic attack of the sulfur atom of glutathione on the electrophilic groups of a wide range of exogenous and endogenous compounds (Probable). Involved in the formation of glutathione conjugates of both prostaglandin A2 (PGA2) and prostaglandin J2 (PGJ2) (PubMed:9084911). It also catalyzes the isomerization of D5-androstene-3,17-dione (AD) into D4-androstene-3,17-dione and may therefore play an important role in hormone biosynthesis (PubMed:11152686). Through its glutathione-dependent peroxidase activity toward the fatty acid hydroperoxide (13S)-hydroperoxy-(9Z,11E)-octadecadienoate/13-HPODE it is also involved in the metabolism of oxidized linoleic acid (PubMed:16624487).

Activity summary

The GSTA1 protein also known as Glutathione S-transferase alpha 1 is an important enzyme in detoxification processes in the body. It has a molecular mass of approximately 25 kDa and expresses highly in the liver but is also seen in other tissues like the kidney and lung. It belongs to the glutathione S-transferase family which plays function in the conjugation of reduced glutathione to a variety of endogenous and exogenous electrophilic compounds.

Biological function summary

The GSTA1 protein contributes significantly to the neutralization of oxidative stress by detoxifying harmful compounds. This enzyme shows a high affinity for certain substrates and serves as a detoxifying agent particularly in the liver safeguarding cells from oxidative damage. GSTA1 exists mostly as a homodimer but can also form heterodimers with other related enzymes enhancing its substrate range and activity.

Pathways

GSTA1 plays a role in metabolic and detoxification pathways essential for cellular protection. It is an important player in the glutathione metabolism pathway where it collaborates with other enzymes like GSTM1 to facilitate detoxification processes. Additionally GSTA1 participates in the pathways leading to the synthesis of mercapturic acids which are important for the excretion of toxic metabolites.

GSTA1 is linked to conditions such as cancer and cardiovascular diseases. Altered expression or polymorphisms in the GSTA1 gene can affect the enzyme's ability to detoxify carcinogens making it significant in cancer risk and progression. The GSTA1 protein also interacts with other enzymes like GSTA4 in the context of oxidative stress which relates to cardiovascular disorder pathogenesis.
