Binds to sialic acid-containing receptors on the cell surface, bringing about the attachment of the virus particle to the cell. This attachment induces virion internalization of about two third of the virus particles through clathrin-dependent endocytosis and about one third through a clathrin- and caveolin-independent pathway. Plays a major role in the determination of host range restriction and virulence. Class I viral fusion protein. Responsible for penetration of the virus into the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane. Low pH in endosomes induces an irreversible conformational change in HA2, releasing the fusion hydrophobic peptide. Activity summary

Hemagglutinin also known as HA is a protein with a mass of approximately 70 kDa. This surface protein expresses mainly on the envelope of influenza viruses. Hemagglutinin plays an important role in binding the virus to host cell receptors and facilitates entry of viral particles into the target cells. The protein undergoes a conformational change to promote fusion between the viral envelope and host cell membranes which is essential for viral infectivity.

Biological function summary

Hemagglutinin mediates host immune recognition since it serves as a major antigenic component. The protein forms a trimeric complex with each subunit comprising two functional regions HA1 and HA2. HA1 is responsible for receptor binding while HA2 is involved in membrane fusion. The immunogenic nature of hemagglutinin challenges the immune system to produce antibodies targeting this structure making it a focus for vaccine design.

Pathways

Hemagglutinin directly interacts with the endocytosis pathway. This interaction facilitates the internalization of the virus into endosomes where the acidic environment triggers hemagglutinin-mediated fusion of viral and endosomal membranes. Hemagglutinin's function in these pathways closely relates to viral neuraminidase which aids in the release of progeny viruses from infected host cells after replication.

Hemagglutinin is notably central to influenza infection an acute respiratory condition with significant morbidity and mortality. Antigenic variations in hemagglutinin contribute to seasonal flu epidemics and potential pandemics. These variations also complicate vaccine formulation as antibodies generated against one hemagglutinin variant may be less effective against others. Additionally hemagglutinin maintains a relationship with the M2 ion channel protein which is important in viral uncoating and replication processes thereby influencing virus pathogenicity and disease outcomes.
