Western blot - HRP Anti-TAF9 antibody [EPR12075(B)] (AB209310)

This blot was produced using a 4-12% Bis-tris gel under the MES buffer system. The gel was run at 200V for 35 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab209310 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.

TAF9 also known as TATA-box binding protein-associated factor 9 is a component of the transcription factor IID (TFIID) complex that plays an essential role in the initiation of gene transcription by RNA polymerase II. This protein has a molecular mass of around 28 kDa and is primarily located in the nucleus of the cell. TAF9 is expressed in various tissues with notable expression levels in liver and kidney.

The TFIID basal transcription factor complex plays a major role in the initiation of RNA polymerase II (Pol II)-dependent transcription (PubMed : 33795473). TFIID recognizes and binds promoters with or without a TATA box via its subunit TBP, a TATA-box-binding protein, and promotes assembly of the pre-initiation complex (PIC) (PubMed : 33795473). The TFIID complex consists of TBP and TBP-associated factors (TAFs), including TAF1, TAF2, TAF3, TAF4, TAF5, TAF6, TAF7, TAF8, TAF9, TAF10, TAF11, TAF12 and TAF13 (PubMed : 33795473). TAF9 is also a component of the TBP-free TAFII complex (TFTC), the PCAF histone acetylase complex and the STAGA transcription coactivator-HAT complex (PubMed : 15899866).
