Function

Actin filament (F-actin)-binding protein involved in cell-matrix adhesion and cell-cell adhesion. Regulates cell-surface E-cadherin expression and potentiates mechanosensing by the E-cadherin complex. May also play important roles in cell morphology and locomotion.

    Western blot - HRP Anti-Vinculin antibody [EPR8185] - Loading Control (ab323345)

Vinculin Western blot staining using rabbit Anti-Vinculin antibody

Blocking and diluting buffer and concentration: 5% NFDM/TBST.

    Performed under reducing conditions.

In Western blot, ab323345 was shown to bind specifically to Vinculin. Target of interest was observed at 123 kDa in wild-type Hela cell lysates (lane 1) with no signal observed at this size in Vinculin knockout cell line (lane 2) (lane 2, knockout cell line Human VCL (Vinculin) knockout HeLa cell line ab265580.

In Western blot, Anti-GAPDH antibody [EPR16891] - Loading Control (Anti-GAPDH antibody [EPR16891] - Loading Control ab181602) staining at 1/200000 dilution.

All lanes: Western blot - HRP Anti-Vinculin antibody [EPR8185] - Loading Control (ab323345) at 1/1000 dilution

Lane 1: Wild-type HeLa (human cervical adenocarcinoma epithelial cell) whole cell lysate at 20 µg

Lane 2: Vincin knockout HeLa whole cell lysate at 20 µg

Lane 3: NIH/3T3 (mouse embryonic fibroblast) whole cell lysate at 20 µg

Lane 4: C6 (rat glial tumor glial cell) whole cell lysate at 20 µg

    Performed under reducing conditions.

Observed band size: 123 kDa, 36 kDa

    Exposure time: 3.25s
