The Hsp70l also known as Heat Shock Protein 70-like is a molecular chaperone with an estimated mass of around 70 kDa. It plays a role in protein folding prevention of protein aggregation and assistance in protein transport across cellular membranes. Hsp70l expressions occur mainly in eukaryotic cells especially in response to cellular stress conditions. The presence of Hsp70l in various cellular compartments including the cytoplasm and organelles supports a range of cellular functions to maintain proteostasis.
Biological function summary

The cellular functions and stress response mechanisms insert Hsp70l in various biological processes. It often associates with the Hsp90 complex to assist in the maturation of client proteins. This protein complex facilitates the stabilization and activation of a wide range of substrates including kinases and transcription factors. The participation of Hsp70l in these complexes highlights its role in cellular homeostasis and stress adaptation mechanisms.

Pathways

Hsp70l plays critical roles in cellular stress response and protein repair pathways. An essential pathway includes the unfolded protein response pathway where Hsp70l contributes to refolding damaged proteins or targeting them for degradation. Additionally Hsp70l interacts with proteins like Hip and Hop which modulate its activity within these pathways. Through these interactions Hsp70l supports cellular resilience to environmental and physiological stress.

Hsp70l links significantly to neurodegenerative diseases and certain cancers. In neurodegenerative conditions Hsp70l associates with proteins like tau and alpha-synuclein aiding in their proper folding and reducing their aggregation. In cancer increased Hsp70l levels have associations with enhanced tumor cell survival and resistance to chemotherapy.
