HSV2 ICP4 also known as Infected Cell Polypeptide 4 is a significant transcriptional regulator in Herpes Simplex Virus type 2. It works as a major transcription factor necessary for virus replication. ICP4 weighs approximately 160 kDa. It expresses abundantly in cells infected by HSV2 leading to changes in cell function necessary for viral replication. ICP4 binds to DNA in a sequence-specific manner and activates transcription of most viral genes.
Biological function summary

ICP4 is essential to switch on the expression of viral genes during the lytic phase. It is not part of a complex but acts by binding directly to the viral DNA. The protein's function influences the viral lifecycle driving progression into the productive phase. By regulating gene expression ICP4 ensures viral proteins are synthesized in a timely manner during infection.

Pathways

ICP4 plays an instrumental role in the transcriptional activation of HSV2’s gene expression cascade. This pathway includes the viral immediate early (IE) early (E) and late (L) genes ensuring cycle progression. Besides ICP4 regulates interactions with other viral proteins like ICP0 and ICP27 which help maintain efficient replication and transcription processes in infected cells.

ICP4 is closely associated with conditions caused by HSV2 such as genital herpes and its complications.
