The HSV2 ICP8 Major DNA binding protein also known as infected cell protein 8 or ICP8 plays an important role in the replication machinery of the herpes simplex virus type 2 (HSV-2). ICP8 is a single-stranded DNA-binding protein with an approximate mass of 128 kDa. It expresses in large quantities in cells infected by HSV-2 facilitating the assembly of other viral proteins needed for effective viral DNA replication. The protein's structure allows it to bind DNA strongly and support the viral DNA replication process.
Biological function summary

ICP8 interacts with other viral and cellular proteins to organize the replication compartment inside the host cell nucleus. It forms part of the replication complex working alongside HSV-2 DNA polymerase and other accessory proteins to effectively duplicate the viral genome. Participates in the opening of the viral DNA origin to initiate replication by interacting with the origin-binding protein. May disrupt loops, hairpins and other secondary structures present on ssDNA to reduce and eliminate pausing of viral DNA polymerase at specific sites during elongation. Promotes viral DNA recombination by performing strand-transfer, characterized by the ability to transfer a DNA strand from a linear duplex to a complementary single-stranded DNA circle. Can also catalyze the renaturation of complementary single strands. Additionally, reorganizes the host cell nucleus, leading to the formation of prereplicative sites and replication compartments. This process is driven by the protein which can form double-helical filaments in the absence of DNA.
