HtrA3 also known as high temperature requirement A3 is a serine protease that regulates protein quality control by degrading misfolded or damaged proteins. With a molecular mass of approximately 50 kDa HtrA3 is expressed in a variety of tissues including placenta lung and heart. Expression levels vary in different cell types which indicates a diverse role. The active form of HtrA3 often results from the removal of its N-terminal inhibitory domain allowing it to target specific substrates for degradation.
Biological function summary

HtrA3 plays a role in cellular stress response and apoptosis regulation. This protein engages in mitochondrial pathways to exert its pro-apoptotic effects. HtrA3 often functions independently rather than as part of a complex although its influence on pathways can involve interactions with other proteins pivotal to cell survival. In cellular stress conditions HtrA3 modulates the stability and function of key proteins involved in apoptosis.

Serine protease that cleaves beta-casein/CSN2 as well as several extracellular matrix (ECM) proteoglycans such as decorin/DCN, biglycan/BGN and fibronectin/FN1. Inhibits signaling mediated by TGF-beta family proteins possibly indirectly by degradation of these ECM proteoglycans (By similarity). May act as a tumor suppressor. Negatively regulates, in vitro, trophoblast invasion during placental development and may be involved in the development of the placenta in vivo. May also have a role in ovarian development, granulosa cell differentiation and luteinization (PubMed : 21321049, PubMed : 22229724).
