HuC/HuD proteins often referred to as HuC/D markers or HuC protein are part of the ELAV (embryonic lethal abnormal visual system) family. These proteins are RNA-binding and weigh approximately 40kDa. They show expression mainly in neuronal tissues acting as post-transcriptional regulators within neurons. HuC/D proteins are used as markers for neural differentiation particularly in studies involving developing neural tissues or cultured neurons such as those in HuC zebrafish models.
Biological function summary

These RNA-binding proteins regulate mRNA stability and translation thereby influencing neuronal differentiation and function. HuC/D proteins do not operate as isolated units; instead they form complexes with various RNA molecules. These complexes facilitate the stabilization and transport of mRNAn important for the synthesis of certain neuronal proteins. Combined activity of HuC and HuD is significant in maintaining cellular identity in neurons.

Pathways

HuC/D proteins play roles in neuronal differentiation and maintenance pathways. One of these pathways is the MAPK/ERK pathway which is involved in transmitting chemical signals from the cell surface to the DNA in the cell nucleus and affects neuron survival and growth. The function of HuC/D within this pathway connects them to other proteins like NeuroD and N-myc which are related in the regulation of neural gene expression.

HuC/D proteins have associations with neurodegenerative conditions such as Alzheimer’s disease and Parkinson’s disease. The improper regulation of RNA-binding proteins like HuC/D can lead to neuronal cell death and impaired nervous system function. There is also a connection to paraneoplastic neurological syndromes where the autoimmune response against HuC/D proteins among others leads to neuronal damage. The relationship between HuD and proteins such as TAU becomes relevant here as disruptions in HuD function can impact TAU pathology critical in neurodegenerative diseases.
